Background

TLR6 (toll-like receptor 6, also called CD286) belongs the family of TLRs which are membrane receptors with potential of acting as gatekeepers of the innate immunity through recognizing microbial components and initiating activation of an adequate immune response. Localized in cell membrane as well as in cytoplasmic vesicle/phagosome membrane as single-pass type I membrane protein, TLR6 is expressed in monocytes, CD11c+ immature dendritic cells, plasmacytoid pre-dendritic cells and dermal microvessel endothelial cells. TLR6 participates in the innate immune response to Gram-+ve bacteria and fungi. It specifically recognize diacylated/triacylated lipopeptides, and has the ability to recognize mycoplasmal macrophage-activating lipopeptide-2kD (MALP-2), soluble tuberculosis factor, phenol-soluble modulin and Borrelia burgdorferi outer surface protein A lipoprotein (OspA-L) cooperatively with TLR2. It acts via MYD88 and TRAF6, leading to NFkB activation, cytokine secretion and inflammatory response. Moreover, it has been shown to bind MYD88 and dimerize with TLR2, wherein TLR6- TLR2 heterodimers recognize a large number of lipidcontaining molecules and transducers inflammatory signalling in a variety of cell type.
