Background

p57Kip2 protein is a inhibitor of several G1 cyclin complexes and is a negative regulator of cell proliferation. The gene encoding human p57Kip2 protein is located on chromosome 11p15.5, a region implicated in both sporadic cancers, Wilms tumor and Beckwith-Wiedemann syndrome. Its binding is cyclin dependent. Overexpression of the protein arrests cells in G1. KIP2 mRNA displays a striking pattern of expression during development, showing high level expression in skeletal muscle, brain, heart, lungs, and eye. Most of the KIP2-expressing cells are terminally differentiated, suggesting that it is involved in decisions to exit the cell cycle during development and differentiation. It is located at 11p15.5, a region implicated in sporadic cancers and Beckwith-Wiedemann syndrome, P57 kip2 is a familial cancer syndrome, marking it as a candidate tumor suppressor. Alternate splicing of this gene generates several transcript variants encoding two different isoforms.
