Background

Metabotropic Glutamate Receptor GRM8 binds L-glutamate, the major excitatory neurotransmitter in the central nervous system, which activates both ionotropic and metabotropic glutamate receptors. Glutamatergic neurotransmission is involved in most aspects of normal brain function and can be perturbed in many neuropathologic conditions. Based on sequence homology, putative signal transduction mechanisms, and pharmacologic properties, GRM8 has been placed in Group III along with GRM4, GRM6, and GRM7. Group II and Group III receptors are linked to inhibition of the cyclic AMP signaling cascade, but differ in their agonist selectivities. Three isoforms of GRM8 are produced by alternative splicing. GRM8 has been reported primarily in the brain. ESTs have been isolated from B-cell/lung/testis, brain, breast, kidney, and testis libraries.
