Background

NSD family of SET domain-containing histone methyltransferases includes NSD1, NSD2 (also known as WHSC1, Wolf-Hirschhorn syndrome candidate 1 or MMSET, multiple myeloma SET), and NSD3 (also known as WHSC1L1, WHSC1 like 1). All the three NSD proteins have been directly linked to multiple human diseases and each NSD family member behaves as an oncogene. HSD3 is a istone methyltransferase that preferentially methylates Lys-4 and Lys-27 of histone H3. H3 Lys-4 methylation represents a specific tag for epigenetic transcriptional activation, while Lys-27 is a mark for transcriptional repression. Translocations resulting in NUP98 fusion to NSD1 and NSD3 leads to the development of acute myeloid leukemia (AML) and overexpression of NSD3 via gene amplification occurs in several breast cancer cell lines as well as in non small cell lung carcinomas (NSCLC).
