Type I membrane protein. Localizes to late endocytic compartment. Associates with lysosome membranes.
Specificity
NB100-65011 recognizes a polymorphic determinant of rat Ia antigen (RT1Bu) present on Lewis, Wistar and AO strain rats but not BN, DA or PVG/c strains (1,2). This antibody is useful for distinguishing Ia positive cells from different rat strains, e.g. for recognizing cells of donor origin in bone marrow reconstituted radiation chimaeras (3,4). MRC OX-3 cross reacts with mouse strains of MHC haplotypes b and s, and analysis of recombinant mouse strains showed that the determinants mapped to the I-A region. NB100-65011 recognizes Ia antigens on B-cells, dendritic cells and certain epithelial cells (1,3,4). This product is routinely tested in flow cytometry on Lewis rat splenocytes.

Background

MHC Class II antigens are heterodimers consisting of one alpha chain(31-34kD) and one beta chain (26-29kD). The family of monoclonal antibodies (ER-TR 3, ER-TR 2, ER-TR 1) detect MHC class II antigens encoded by the murine Ia region of the H-2 complex, corresponding to the human HLA-DR region. MHC Class II antigens are a valuable tool for studying T helper cell interaction with class II positive antigen presenting cells (dendritic cells, B cells, macrophages) and offer new possibilities for studying the development of T helper cells since these antibodies also stain stromal cells in the thymus. MHC Class II antigens are also inducible on a number of other cells (endothelium and epithelial cells) by interferon gamma.
