L-Selectin/CD62L Antibody (FMC46) [FITC] Summary

Immunogen
PHA stimulated lymphoblasts.
Localization
Membrane; single pass type I membrane protein.
Specificity
NB100-65388 recognizes human CD62L, also known a L-selectin, a 74-95kDa member of the selectin family of adhesion receptors, which acts as a ligand for both CD62-P (P-selectin) and CD62E (E-selectin). Human CD62L is constitutively expressed on most leucocytes including monocytes, granulocytes, lymphocytes, NK cells, bone marrow myeloid progenitor cells and on a subset of thymocytes. CD62L plays an important role in leucocyte tethering and rolling on the endothelial cell surface and for the homing of naive lymphocytes to lymph nodes and Peyers patches via HEV. Neutrophils require a constant supply of this molecule on the cell surface for migration into peripheral tissues and adhesion to activated endothelium at sites of inflammation, where CD62L is rapidly shed as soluble L-selectin, but surface expression still remains. The expression of CD62L is down regulated on lymphocytes and neutrophils by PMA stimulation.

Background

The human CD62L is a 74-95 kDa glycoprotein member of the selectin family of adhesion receptors. Human CD62L (L Selectin) is comprised of an aminoterminal C type lectin binding domain, an epidermal growth factor like domain, two short consensus repeat (SCR) sequences homologous to those found in complement binding proteins, a short spacer region, a transmembrane region and a short cytoplasmic region. Human CD62L is constitutively expressed on all classes of leukocytes including lymphocytes (except a substantial population of memory T cells), monocytes and polymorphonuclear cells. It is expressed on bone marrow myeloid progenitor cells, erythroid precursor cells and some thymocytes. It is preferentially localized to the tips of lymphocytes and polymorphonuclear cells microvilli. Bioactive soluble L Selectin derived from lymphocytes and neutrophils is present in normal and pathological biological fluids.
