Applications/Dilutions

Dilutions
  • Western Blot 1:1000
  • Immunohistochemistry 1:10-1:500
  • Immunohistochemistry-Paraffin 1:10-1:500
Application Notes
This antibody is useful in Immunohistochemistry and Western Blot. For WB use at a dilution of 1:1000 (human and bovine) or 1:5000 (mouse and rat). 1:5000 dilution was sufficient to detect 50ng of recombinant rat protein. Detects a band of approximately 58 kDa (predicted molecular weight: 42 kDa).

Reactivity Notes

Cross-reacts with Human, Mouse, Rat and Cow. Whilst reactivity with human and bovine protein is detected, this reactivity is much weaker than that observed with samples from rat or mouse origin. Not yet tested in other species.

Background

Hip (Hsc70-interacting protein) is a cytosolic protein that exists as a homo-oligomer and participates in the regulation of Hsc70 in eukaryotic cells. Hip interacts with the ATPase domain of Hsc70 molecules through its tetratricopeptide repeats and flanking charged alpha-helices. Upon binding to Hsc70, Hip stabilizes the ADP-bound state of Hsc70, a conformation that has a high affinity for unfolded substrate proteins. Together with Hsp40 and BAG-1, Hip may regulate the chaperone activity of Hsc70 by stabilizing the chaperone-substrate complex.
