Background

GPR17, a former orphan G-coupled protein is specifically activated by P2Y and CysLT endogenous ligands, leading to both adenylyl cyclase inhibition and intracellular calcium increases. In both rats and humans GPR17 expressed in the organs which suffer ischemic damage, like the brain, heart and kidneys. Studies by Ciana, et. al. have shown that inhibition of GPR17 reduces ischemic damage in a rat focal ischemia model. These results suggest GPR17 is the common molecular target mediating brain damage by nucleotides and CysLTs.
