Background

Fibrillarin (FBL; human homologue of yeast Nop1p) is the most abundant proteins in fibrillar regions of the eukaryotic cell nucleolus, the site for early phase of pre-rRNA processing, and is considered an excellent marker for nucleolus. FBL interacts with several proteins such as NOLC1, DDX5, C1QBP, PRMT5 and UTP20; and with U3, U8, U13, X and Y small nuclear RNAs also. FBL forms a component of several ribosomal/nucleolar protein complexes and of snoRNP (box C/D small nucleolar ribonucleoprotein) particles that contain NHP2L1, FBL, NOP5 and NOP56, plus a guide RNA. In pre-ribosomal RNA, FBL utilizes the methyl donor S-adenosyl-L-methionine to catalyze the site-specific 2-hydroxyl methylation of ribose moieties, the site specificity being offered by the guide RNA that base pairs with the substrate, and the methylation occurs at a unique distance from the sequence involved in base pairing with the guide RNA. In humans, FBL also acts as a nucleolar autoantigen for non-hereditary immune disease scleroderma, a disease in which the patients have strong circulating FBL autoantibodies.
