Background

Cytokeratin 18 (CK-18), a normal constituent of the hepatocyte cytoskeleton) is a standard tumor marker. CK-18 is elevated in patients with non-malignant liver diseases, particularly in those with prominent cytolysis (1). CK-18 and the homologous mouse Endo B are type I IF protein subunits whose expression is restricted in adults to a variety of simple epithelial tissues. Keratin 8 and 18 (CK8/18) are the major keratin pair in simple-type epithelia, as found in the liver, pancreas, and intestine. Transgenic mice that express mutant keratin 18 develop chronic hepatitis, and have an increased susceptibility to drug-induced hepatotoxicity (2). Studies shows that high CK18 expression in tumor cells is associated with reduced invasiveness in vitro and lack of tumorigenicity in nude mice. It has been shown that high K18 expression correlated with a good prognosis and that reducing K18 expression increased the aggressiveness of established breast cancer cell lines (3).
